Duality of longevity drug explained

Friday, March 30, 2012

A Penn- and MIT-led team explained how rapamycin, a drug that extends mouse lifespan, also causes insulin resistance. The researchers showed in an animal model that they could, in principle, separate the effects, which depend on inhibiting two protein complexes, mTORC1 and mTORC2, respectively.

The study suggests that molecules that specifically inhibit mTORC1 may combat age-related diseases without the insulin-resistance side effect, which can predispose people to diabetes.

Senior author Joseph A. Baur, PhD, assistant professor of Physiology, Perelman School of Medicine, University of Pennsylvania, and colleagues at the Whitehead Institute for Biomedical Research and Broad Institute, Massachusetts Institute of Technology, in Cambridge, MA, describe their work in this week's issue of Science. Baur is also a member of Penn's Institute for Diabetes, Obesity, and Metabolism.

"The hope is that in the future, we will be able to develop molecules that target mTORC1 specifically, separating out the beneficial effects of rapamycin on aging and disease, and leaving behind the insulin-resistance side effect," says Baur.

"Our results demonstrate that reduced mTORC1 signaling is sufficient to extend lifespan and mTORC2 signaling has profound effects on metabolism," says co-first author Lan Ye, PhD, postdoctoral fellow in the Baur lab. "Our findings indicate that mTORC2 may be an important player in the pathogenesis of type 2 diabetes and metabolic syndrome."

One Compound, Many Effects

Rapamycin extends the lifespan of yeast, flies, and mice and is also an immunosuppressant drug for organ transplants and an anti-cancer drug. It was first discovered as a byproduct of Streptomycin hygroscopicus, a bacterium found in a soil sample from Easter Island, an island also known as Rapa Nui, hence the name. Rapamycin was originally developed as an antifungal agent, but that use was abandoned when it was discovered to have immunosuppressive properties.

The mTOR complexes, for mammalian (or mechanistic) target of rapamycin, are proteins that regulate cell growth, movement, and survival, as well as protein synthesis and transcription. Specifically, there are two mTOR complexes and one mTOR protein. The mTOR protein is the core of both complexes (mTORC1 and mTORC2), which behave differently based on their associated proteins. One or both of the mTOR complexes can be inappropriately activated in certain cancers, and dual-specific inhibitors are being developed as chemotherapeutic agents.

Several theories have been put forward by researchers to explain the observations that patients receiving rapamycin are more prone to developing glucose intolerance, which can lead to diabetes. Chronic treatment with rapamycin impairs glucose metabolism and the correct functioning of insulin in mice, despite extending lifespan. The research team demonstrated that rapamycin disrupts mTORC2 in the mice, and that mTORC2 is required for the insulin-mediated suppression of glucose metabolism in the liver.

On the other hand, they also demonstrated that decreasing mTORC1 signaling was sufficient to extend lifespan independently from changes in glucose metabolism. They used a mouse strain in which mTORC1 activity was decreased and saw that lifespan was extended by 14 percent, yet the animals had normal glucose metabolism and insulin sensitivity.

"Besides developing more specific inhibitors for mTORC1, we remain very interested in understanding why mTORC1 inhibition extends life in the first place," explains Baur. "We're currently looking at the interactions between mTORC1 and other pathways that influence longevity, as well as its effects on things like free radical generation and protein quality control."

The MIT colleagues on the Science paper are co-first author Dudley W. Lamming, PhD, Whitehead Institute, and co-senior author David M. Sabatini, MD, PhD, member of Whitehead Institute, Howard Hughes Medical Institute investigator, and professor of biology at MIT. This study was funded by grants from the American Federation for Aging Research, the National Cancer Institute, and the Bingham Trust, through Penn's Institute on Aging.

Some factors to consider when selecting from the different loan programs. Your current financial situation, would you expect this situation to change? How comfortable are you using a changing mortgage payment? A fixed interest rate mortgage can save you thousands in interest in the period of the loan, but it will likely give you higher regular monthly mortgage rates. An adjustable charge will start you out along with lower monthly payments however you could face increased monthly payments if the rates change.

You should also consider using a professional similar to Francopee finance advice help you obtain control of your loan or financial debt long term.

You have decided which type of loan is best for an individual, now you need to select which of the more popular home loan programs, is the best one for you.

Typical loans are guaranteed by government backed lenders. They are also referred to as government sponsored agencies (GSEs). They can be used to purchase in order to refinance single household or 4 plex residences with a first or perhaps a second mortgage. There are generally limits that are fine-tuned annually if needed in line with the national average of recent homes. You would need to verify what the current years limitations are for an precise amount if you were to select this type of home loan software.

FHA loans help low income families become home owners. By protecting a mortgage company from default they encourage companies to make loans in order to families that many certainly not meet normal credit rating guidelines. Some of the highlights of these loans are. Lower down payments can be as lower a 3% versus the conventional 10% requirements. Closing costs of up to 2 or 3 per cent of the house value can be funded, this reduces the at the start money needed. The particular FHA also imposes limitations on the fees through the mortgage company such as the mortgage origination fee can not be more than 1% of the quantity of your mortgage.

You should also consider using a professional similar to FRMVB Financial Advice Service help you acquire control of their finance or financial debt long term.

VA loans are available to armed service veterans who were on active military work and were released under conditions besides dishonourable discharge. Veterans with services only during peacetime times and active obligation military personnel have to have had more than One hundred eighty days of active service. There are other qualification requirements. If you think you might be eligible contact your nearby or state veterans government representative.

The biggest factor in a VA mortgage loan is that no advance payment is required in most cases. There is not any mortgage insurance installments needed, closing costs for the buyer are also limited. You can negotiate charges with the lender and you then have a choice of payment plans with as much as a 30 year bank loan.

You should also consider employing a professional like Ignite Salem Personal Finance help you acquire control of your finance or debt long term.

The last loan plan we will mention is known as a subprime loan. This is a mortgage for people with poor credit who not qualify for a normal loan or a VA or FHA confirmed loan. These loans generally will require a larger advance payment and have a higher interest. This is because of the risk involved to the mortgage company. These lending options should normally be looked at for a limited period of time such as 2 to 4 years. It is a good way to boost your credit score situation and then remortgage with more favorable conditions.

We have shown discovering or planning that new dream residence is just the beginning of the quest into your new home. The proper answer to the question, which of the home loan programs is for you, takes research and a trustworthy look at your personal situation.

(AP) ? Streets of Damascus that were crowded until late at night stand mostly empty after sunset as Syrians stay home, fearing robberies and kidnapping. After a year of chaos, a country that once boasted it was the safest in the Middle East is experiencing a surge in crime.

Reports of thefts, armed robberies and house break-ins have swelled, an apparent sign of criminals taking advantage of the turmoil as the regime of President Bashar Assad wages a bloody, nationwide crackdown against an opposition demanding his ouster.

The government blames members of the opposition or army defectors for much of the crime. The opposition, in turn, blames the regime, saying it is allowing the lawlessness so it can tell the public that the uprising has brought chaos.

But it may be that security forces are now too distracted. Syria's 17 security and intelligence agencies used to control the country with an iron fist, and crime was rare as many feared the regime's brutality. Several pardons issued by Assad over the past year as a gesture to the opposition are believed to have freed criminals ? including convicted murderers and thieves ? alongside some political prisoners.

Whatever the cause, Damascus residents say criminals are operating with unprecedented boldness.

A woman who lives in the posh Damascus neighborhood of Tijara recounted a harrowing armed robbery against her family that she said took place earlier this month. At 6 in the evening, three gunmen with automatic weapons banged on their apartment door while her mother and 8-year-old brother were home alone. The brother opened the door and the assailants stormed in, she told The Associated Press by phone from the Syrian capital.

The gunmen held a gun to the boy's head and told the mother they would shoot unless she handed over her jewelry and cash, the woman said. Her mother gave them all her jewelry and cash worth about $30,000. The gunmen forced the mother and son into a bedroom, tied their hands, locked them in and then fled, said the woman.

"This is the first such case in this neighborhood in 15 years," she said the police told her. The woman spoke on condition of anonymity for fear the criminals might harm members of her family.

Another Damascus resident said a neighbor ? a wealthy, pro-government businessman in his late 40s ? was kidnapped while going to work and was only released after his family paid a ransom worth $300,000. After the December kidnapping the man left Syria along with his wife and children. The resident also spoke on condition of anonymity for fear of making herself a target.

In the central city of Homs, a resident told the AP that gunmen knocked on the door of his neighbor, put a pistol in his face and said to him, "Give us the keys of your car."

The man said the gunmen drove away. The Homs resident, who also spoke on condition of anonymity, said his neighbor still lives in fear.

Damascus residents say that many people who own luxury cars keep them at home and move around in taxis in order for them not to have their cars stolen.

Homs resident Wael, 27, said crime has doubled in some restive neighborhoods because "there is no security and people are now behaving as they want."

"In the past, we used to sleep with our windows open. Now, we cannot do that," said Wael, who asked that he only be identified with his first name.

Iyad al-Bunni, 58, a merchant from Damascus, said the absence of stability and security has allowed thieves to thrive. "Armed groups are using the security vacuum to carry out acts of looting," he said.

Pinning down the extent of the rise in crime is difficult, in part because of the potential for violence connected to the uprising and the regime's crackdown to get mixed in with nonpolitical crime. The regime denies the existence of an uprising, saying it faces not a popular revolt but violence from "terrorists" ? so it likely includes deaths connected to political violence in its crime rate.

The revolt began a year ago with peaceful protests against Assad around the country that were met with gunfire.

Since then, violence has spiraled. Some rebels have taken up arms, attacking army positions and reportedly gunning down pro-regime figures. Security forces have besieged entire cities or rebellious neighborhoods, hammering residential areas with artillery. Pro-regime militiamen known as Shabiha have targeted suspected opposition supporters. Tit-for-tat sectarian killings have taken place between majority Sunnis, who largely back the opposition, and minority Alawites, who stand by the regime.

The United Nations estimates that 8,000 people have been killed in the violence surrounding the uprising.

The Damascus general prosecutor, Marwan Louji, told the pro-government daily Baladana in January that "political, terrorist and normal" crime in the capital doubled in 2011 compared to the previous year. He said theft shot up between 50 to 100 percent. He did not otherwise elaborate.

It was not clear whether his figures included deaths connected to the uprising and crackdown or not. Louji refused an AP request to be interviewed by telephone, and Syrian security officials refused comment.

Louji told Baladana that authorities will work on making sentences harsher in an attempt to reduce crime rates. He gave an example that if a crime is usually punished with a prison sentence ranging between 3 to 15 years in jail, judges will give the maximum punishment.

Mustafa Osso, a Syria-based activist and lawyer who defends political prisoners, blamed the regime for the crimes in order to show that it must stay in power for Syria to be stable.

"They want to say that we either stay in power or fear and chaos will spread in the country," Osso said. "The regime is helping in the rise of crime rates. They released criminals serving long sentences, we are talking about dangerous criminals who committed major crimes."

Mysterious cloud spotted on Mars

Wayne Jaeschke

Amateur astrophotographer Wayne Jaeschke captured this image of a "terminator projection" rising up from the edge of the Martian disk at about the 1 o'clock position on March 22. The inset photo is a 200 percent enlargement of the region around the projection. For more, check out Exosky.net, Jaeschke's website.

By Alan Boyle

Amateur astronomers are puzzling over a seemingly anomalous cloud that has shown up on images of Mars taken over the past few days. Is it really a cloud, or a trick of the eye? Does it really extend 150 miles up from the surface, as some of the observers suggest? And what churned up all that stuff, anyway? The amateurs and the pros will be trying to resolve those questions before the phenomenon fades away.

"It's not completely unexpected," Jonathon Hill, a member of the team at the Mars Space Flight Facility at Arizona State University, told me today. "But it's bigger than we would expect, and it's definitely something that our atmosphere guys want to take a look at."

Hill and his colleagues will be looking at the area where the cloud was spotted using the Thermal Emission Imaging System, or THEMIS, which is one of the instruments on NASA's Mars Odyssey orbiter.

"In the command upload we're preparing to send today, we've included observations that will hopefully capture some of these recent clouds," Hill wrote in an email. "Our THEMIS camera on Mars Odyssey is capable of acquiring simultaneous visible and thermal infrared images, so our atmospheric researchers are pretty excited about the possibility of not only getting a good look at the cloud structures, but also their temperatures."

THEMIS will be checking out heightened cloud activity around Mars' shield volcanoes as well as around the southern site spotted by the amateurs. Pictures from a camera on NASA's Mars Reconnaissance Orbiter, called the Mars Color Imager, or MARCI, might provide further clues about the southern cloud feature. And amateur astronomers are sending out the alert for observers to keep a close watch on the Red Planet over the coming days.

There's been lots of buzz about the high-altitude cloud on Cloudy Nights and other online discussion forums for skywatchers. Sky & Telescope's Sean Walker says the puff of white was first noticed on March 20 by Wayne Jaeschke, an amateur astrophotographer from Pennsylvania. Since then, other observers have identified the feature in images going back as far as March 12.

All sorts of hypotheses have been proposed: Could it be debris kicked up by a meteoric impact? Is it a huge weather system? Is it merely a funny kind of glint caused by a combination of lighting and atmospheric conditions?

In an email, Jaeschke told me that the feature is "still there, although it has decreased in size over the past two days."

"This has led some to believe that it was some sort of transient-type event," Jaeschke said. So it's crucial to make as many observations of the area as possible over the next few days.

Wayne Jaeschke created this animated GIF image of Mars with the cloud coming into view on the upper right edge of the planet's disk. For more from Jaeschke, check out his Exosky website.

Walker says the feature is currently well-placed for viewing from the Americas. He says it should show up on the edge of Mars' disk around 1:10 a.m. ET Saturday, and 39.5 minutes later on each succeeding night. Consult the photos above for guidance on where to point a medium-size telescope ? keeping in mind that these images are inverted to appear as they would through a telescope, with south pointing "up." Arizona State University's Hill says the area in question is called Terra Cimmeria.

Observation reports should be sent via email to Richard McKim, director of the British Astronomical Association's Mars Section. (The linked website includes McKim's email address, as well as a picture of the chap.)

Solving this mystery ? if it indeed turns out to be an honest-to-goodness mystery rather than a mere quirk?? may require additional data from the big guns of the astronomy world. But in any case, the episode illustrates once again how much amateurs can contribute to uncovering the wonders of the cosmos.

"When it comes to Mars, amateurs and professionals working together give you way more insight into ongoing processes, because with so many amateurs, you're continuously monitoring changes in the planet," Hill told me. "They provide a perspective and a context that we don't usually get."

LOS ANGELES (AP) ? Drugs took many things from Whitney Houston ? her pristine voice, clean image and her career ? and coroner's officials revealed Thursday that cocaine also played a role in the Grammy winner's death in the bathtub of a luxury hotel nearly six weeks ago.

Houston drowned accidentally at the Beverly Hilton Hotel on Feb. 11, and autopsy results revealed heart disease and her recent and chronic cocaine use were contributing factors.

The results ended weeks of speculation about what killed the singer-actress at age 48 on the eve of the Grammy Awards and a planned comeback. Instead, Houston now joins the long list of entertainers who have died early with drugs at least partly to blame.

Coroner's officials did not reveal how much cocaine was found in Houston's system, but said there were signs of recent and chronic use. It was unclear whether she suffered a heart attack before drowning, Assistant Chief Coroner Ed Winter said.

"We are saddened to learn of the toxicology results, although we are glad to now have closure," Patricia Houston, the singer's sister-in-law and manager, said in a statement.

Houston's death the night before the Grammys altered the awards ceremony and brought back immediate memories of the singer in her better days, belting out hits and starring in the feature films such as "The Bodyguard" and "Waiting to Exhale." Years later, Houston's drug use had robbed her of her ability to hit high notes and left her once clean image tarnished.

"The biggest devil is me. I'm either my best friend or my worst enemy," Houston told ABC's Diane Sawyer in an infamous 2002 television interview with then-husband Bobby Brown by her side.

In 2009, after she had divorced Brown, she told Oprah Winfrey that her cocaine and marijuana use took over her life. Both drugs were found in Houston's system after she died, though the marijuana and several other prescription drugs played no role in her death, coroner's officials said.

"I had so much money and so much access to what I wanted," Houston told Winfrey. "I didn't think about the singing part anymore. I was looking for my young womanhood."

Houston's planned comeback after that interview didn't pan out, but she was taking another shot this year with her appearance in a remake of the film "Sparkle."

She stars as the mother of a family of girls who form a singing group and struggle with fame and addiction. She also sang for the film, delivering a soulful rendition of the gospel hymn "His Eye is on the Sparrow."

It is unclear what impact, if any, the autopsy results will have on the film, although untimely death has not been an impediment to the posthumous releases of other actors and entertainers. Heath Ledger's performance in "The Dark Knight" helped propel the film to blockbuster status and earned the actor a posthumous Oscar. He died of an accidental prescription drug overdose in January 2008, months before the film's release.

More recently, Amy Winehouse's duet "Body and Soul" with Tony Bennett earned them a Grammy Award, months after the singer died unexpectedly in London. An inquest determined she died from an accidental alcohol overdose.

A spokesman for Sony Pictures did not return an email message Thursday seeking comment on whether Houston's autopsy results would alter marketing of "Sparkle."

It will be a couple weeks before the exact amount of cocaine in Houston's system is released, officials said.

The drug has been known to cause damage to the heart and could have cause Houston's death, said Dr. Michael Fishbein, professor of pathology at the David Geffen School of Medicine at UCLA. He had no role in the investigation.

He said a likely scenario was that Houston's cocaine use interfered with the normal function of her heart.

"There's no reason to drown in a bathtub unless you're incapacitated," Fishbein said.

Houston's friend and collaborators said after her death that they didn't believe she was still abusing drugs, and she described as being a complete professional on the "Sparkle" set.

Houston, a sensation from her first, eponymous album in 1985, was one of the world's best-selling artists from the mid-1980s to the late 1990s, turning out such hits as "I Wanna Dance With Somebody," ''How Will I Know," ''The Greatest Love of All" and "I Will Always Love You."

She was buried last month in a New Jersey cemetery next to her father after an emotional four-hour funeral service that was attended by friends, family and superstars such as Oprah Winfrey, Tyler Perry, Alicia Keys, Mariah Carey, Mary J. Blige, Jennifer Hudson and Roberta Flack

Christian family evicted from Egypt town

CAIRO (AP) ? Nabil Gergis, a Coptic Christian, lived for nearly two decades in the Egyptian town of Amriya, raising his children and managing a modest business. Those ties couldn't protect him after a sex video purportedly showing his brother with a Muslim woman began to circulate.

Angry residents in the conservative, Muslim-majority town held protests and set fire to the Gergis family businesses. None of the attackers was prosecuted. Instead, a committee of tribal elders, local lawmakers and security officials ordered the 11 members of the Gergis family ? the brother, Nabil and others ? to leave town.

The story of Amriya demonstrates one of the reasons Egypt's Coptic Christian minority and even some in the Muslim majority feel the situation is precarious, particularly since the ouster of former President Hosni Mubarak a year ago. The rule of law, they and human rights groups say, is being eclipsed by such "reconciliation councils," trying to fill the security vacuum left by Mubarak's fall.

"There is no law that would have found me responsible for anything, and under the law I would have never been kicked out of my home," said Nabil Gergis. He said he, his wife and their two children do not know who to turn to protect their rights and that he feels the government has turned its back on them.

Egypt's Copts are mourning Pope Shenouda II, who led the Church for 40 years and died on Saturday. "Baba Shenouda," as he was called, was seen by many in the community as their biggest protector in a country where Christians make up about 10 percent of a population of 85 million.

Shenouda's approach was deeply conservative. He was a close ally of Mubarak during the former president's 29-year rule and used his influence behind the scenes to try to ensure some protections for Christians. But he largely resisted any public protests or pressure. His critics among the Coptic community say that left Christians' rights dependent on personal relations rather than enforced by law.

Reconciliation meetings were not unusual under Mubarak's regime. Muslim-Christian violence broke out occasionally in towns of the south, sometimes in local disputes that turned sectarian. Rather than prosecuting those responsible, local leaders and security officials would often insist on negotiated solutions to keep the peace ? or, critics say, because they were reluctant to confront Muslims involved in the incidents.

The Amriya case was unique because the punishment was so extensive. The town is comprised of scattered villages with some 500,000 residents, about 15 percent of them are Christian.

The incident erupted in late January, when the explicit video allegedly showing Nabil Gergis' brother with a Muslim woman circulated on residents' cell phones. The brother, who is married, has denied any affair.

Any sex outside of marriage is a lightning rod for controversy in the Muslim world, where a woman's chastity is vociferously protected by her family. That a Christian man might have an affair with a Muslim woman only further fanned the flames.

The rumors sparked widespread protests by Amriya residents, who are mostly tribal and deeply traditional. Angry residents set fire to three stores owned by the Gergis' family, which were under their homes. Some Muslim residents tried to help, but were outnumbered by the ultraconservative rioters.

Police showed up hours later and instead of investigating the attack called in the brother for questioning, Gergis said.

With tempers still high, local officials and tribal leaders held a series of meetings and decided to order the expulsion of the entire Gergis family. A Muslim family who had fired shots in the air during the protest to protect their property were initially told they must leave too, but were later allowed to return.

Amriya police argued that they could not guarantee the Gergis family's safety in the face of angry protesters, according to security officials and the Gergis family. Last week, with the family gone, their homes were robbed of cash and other belongings they had to leave behind, Gergis said.

An Egyptian rights group that looked into the case, the Egyptian Initiative for Personal Rights, criticized the expulsion, saying the victims were forced to accept "the outcomes of illegal reconciliation processes" and to abandon their rights.

Ishak Ibrahim, a researcher with the group, warns that the breakdown of security since Mubarak's fall could lead to an even greater dependence on such "reconciliation meetings."

"When the state's authority is eroded, then the community turns to its own leaders to solve the crisis," he said. "The problem is that the outcomes are not consistent. It also means that Christians or the side that is weaker will be at a disadvantage, and so this affects all Egyptians."

The unreliability of the law has hurt Christians in other ways. One sore point is the construction of churches, which requires permissions from security officials and was rarely granted. In response, Christians often built churches secretly, and in several instances in recent years Muslim mobs attacked the construction. Again, perpetrators of such attacks were almost never prosecuted.

Since Mubarak's fall, there has been talk of a law putting construction of mosques and churches under equal rules, but no law has been passed.

A series of attacks over the last year has also stoked Christian fears. A year ago, a Muslim-Christian love affair led a Muslim mob to torch a church in the village of Soul, 18 miles (30 kilometers) south of Cairo. Christians protesting the burning were attacked by a mob; 13 people died and 140 injured.

In May, ultraconservative followers of the Salafi trend of Islam burnt a church in the Cairo working-class district of Imbaba and clashed with Christians, leaving 12 people dead. Many of the rioters believed that a Christian woman who fell in love with a Muslim man had converted to Islam and was being held prisoner by the church.

In October, a Cairo protest led by Copts demanding greater rights was crushed by soldiers, leaving 27 people, mostly Copts, dead.

The Gergis family is now living with relatives in Alexandria. Nabil Gergis said neither the police nor government officials have responded to his pleas for help.

"The only thing that this means is that police don't want us in the country," he said. "It's a feeling that I cannot describe. My children don't have a home anymore."

JPMorgan wins dismissal of Ponzi scheme accusations

(Reuters) - A federal appeals court dismissed a lawsuit seeking to hold JPMorgan Chase & Co responsible for services allegedly provided by the former Washington Mutual Inc to advance a Ponzi scheme at Caribbean-based Millennium Bank.

The 9th U.S. Circuit Court of Appeals said on Tuesday that investors who bought fraudulent certificates of deposit from Millennium did not pursue all remedies with the Federal Deposit Insurance Corporation (FDIC) as required by federal law or show that JPMorgan did anything to advance the alleged fraud.

The decision by a three-judge panel upheld an October 2010 lower court ruling. Niall McCarthy, a lawyer for the investors, was not immediately available for comment.

Washington Mutual was seized on September 25, 2008 by the FDIC, which immediately sold the largest U.S. savings and loan's banking operations to JPMorgan for $1.88 billion.

Last month, a San Francisco federal grand jury returned a 23-count indictment charging William Wise and Jacquline Hoegel with fraud and conspiracy over the sale of $129.5 million of CDs by their companies Millennium, United Trust of Switzerland and Sterling Bank and Trust, resulting in $75 million of losses.

Prosecutors alleged that Millennium, United Trust and Sterling had promised CD investors high rates of return, sometimes over 16 percent, based on overseas investments, but that Wise and Hoegel used investors' funds to enrich themselves and pay off earlier investors.

Their activities were shut down in March 2009 when the U.S. Securities and Exchange Commission filed related civil fraud charges.

Wise, a Canadian lawyer, is believed to be in Canada and a warrant for his arrest has been issued, the U.S. Attorney's office in San Francisco said on Tuesday. Hoegel, a resident of American Canyon, California, was released on bond.

The CD investors sought to hold JPMorgan liable for Washington Mutual's having provided banking services to Millennium. These investors claimed that Washington Mutual knew about the fraud at Millennium, which was based in St. Vincent and the Grenadines, while it was providing these services.

But the appeals court said the investors did not fully present their claims to the FDIC before suing the purchaser, in this case, JPMorgan, of a failed bank.

It added that investors did not show a single specific wrongful act by JPMorgan, relying instead on assertions that the bank's general "practices" caused their losses.

"The plaintiffs' conclusory allegations regarding JPMorgan fall short," Judge Carlos Lucero wrote for the appeals court.

The case is Benson et al v. JPMorgan Chase Bank NA, 9th U.S. Circuit Court of Appeals, Nos. 10-17402 and 10-17404.

O'Dwyer extradition raises copyright profile | law, less ordinary ...

As Cameron and Obama were recently discussing the current extradition laws between the 2 countries, Home Secretary Theresa May approved?the extradition of Richard O?Dwyer to the US, after a UK court had earlier decided he could be extradited for copyright infringement for hosting sites that provided links to unauthorised copyright material on other sites, which could then be download.

Mr O?Dwyer argues he had not downloaded the infringing material but merely directed searchers to other sites.

The two countries? copyright infringement laws differ; Mr O?Dwyer is facing a maximum penalty of 5 years imprisonment in the US, compared with 2 years in the UK.

This decision will certainly concern UK infringers; although whether this will set a precedent will perhaps depend on Cameron and Obama?s recent discussions. Either way, we can be sure that Mr O?Dwyer?s case will only add to the current debate?on whether UK copyright law can stand up to technological advances.

Daydreaming Makes You Smarter [Science]

At high school, it's invariably the kids that day dream who get told off. But a new study suggests that it's those of us whose minds wander that have the best working memory—and working memory is itself directly associated to intelligence. More »

Researchers building melanoma vaccine to combat skin cancer

Monday, March 19, 2012

Mayo Clinic researchers have trained mouse immune systems to eradicate skin cancer from within, using a genetic combination of human DNA from melanoma cells and a cousin of the rabies virus. The strategy, called cancer immunotherapy, uses a genetically engineered version of the vesicular stomatitis virus to deliver a broad spectrum of genes derived from melanoma cancer cells directly into tumors. In early studies, 60 percent of tumor-burdened mice were cured in fewer than three months and with minimal side effects. Results of the latest study appear this week in the journal Nature Biotechnology.

"We believe that this new technique will help us to identify a whole new set of genes that encode antigens that are important in stimulating the immune system to reject cancer. In particular, we have seen that several proteins need to be expressed together to generate the most effective rejection of the tumors in mice," says Richard Vile, Ph.D., a Mayo Clinic researcher in the Department of Molecular Medicine and a coauthor of the study, along with Jose Pulido, M.D., a Mayo Clinic ophthalmologist and ocular oncologist.

Dr. Vile's success with melanoma adds to Mayo Clinic's growing portfolio of experimental cancer vaccines, which includes an active clinical trial of vesicular stomatitis vaccines for liver cancers. Future studies could include similar vaccines for more aggressive cancers, such as lung, brain and pancreatic.

"I do believe we can create vaccines that will knock them off one by one," Dr. Vile says. "By vaccinating against multiple proteins at once, we hope that we will be able to treat both the primary tumor and also protect against recurrence."

The immune system functions on a seek-and-destroy platform and has fine-tuned its capacity to identify viral invaders such as vesicular stomatitis virus. Part of the appeal of building cancer vaccines from the whole spectrum of tumor DNA is that tumors can adapt to the repeated attacks of a healthy immune system and display fewer antigens (or signposts) that the immune system can identify.

Cancers can learn to hide from a normal immune system, but appear unable to escape an immune system trained by the vesicular stomatitis virus with the wide range of DNA used in the library approach.

"Nobody knows how many antigens the immune system can really see on tumor cells," says Dr. Vile. "By expressing all of these proteins in highly immunogenic viruses, we increased their visibility to the immune system. The immune system now thinks it is being invaded by the viruses, which are expressing cancer-related antigens that should be eliminated."

Much immunotherapy research has slowed because of researchers' inability to isolate a sufficiently diverse collection of antigens in tumor cells. Tumors in these scenarios are able to mutate and reestablish themselves in spite of the body's immune system.

The study was a Mayo collaboration with professors Alan Melcher and Peter Selby at the Leeds Institute of Molecular Medicine, University of Leeds, U.K. They were also co-authors.

Church Puts Legal Pressure on Abuse Victims' Group - Laurie ...

Thanks to potteryshard for the link

alt text
David Clohessy keeps records from his advocacy group, known as SNAP, in his St. Louis home.

Turning the tables on an advocacy group that has long supported victims of pedophile priests, lawyers for the Roman Catholic Church and priests accused of sexual abuse in two Missouri cases have gone to court to compel the group to disclose more than two decades of e-mails that could include correspondence with victims, lawyers, whistle-blowers, witnesses, the police, prosecutors and journalists.

The group, the Survivors Network of those Abused by Priests, known as SNAP, is neither a plaintiff nor a defendant in the litigation. But the group has been subpoenaed five times in recent months in Kansas City and St. Louis, and its national director, David Clohessy, was questioned by a battery of lawyers for more than six hours this year. A judge in Kansas City ruled that the network must comply because it ?almost certainly? had information relevant to the case.

The network and its allies say the legal action is part of a campaign by the church to cripple an organization that has been the most visible defender of victims, and a relentless adversary, for more than two decades. ?If there is one group that the higher-ups, the bishops, would like to see silenced,? said Marci A. Hamilton, a law professor at Yeshiva University and an advocate for victims of clergy sex crimes, ?it definitely would be SNAP. And that?s what they?re going after. They?re trying to find a way to silence SNAP.?

Lawyers for the church and priests say they cannot comment because of a judge?s order. But William Donohue, president of the Catholic League for Religious and Civil Rights, a church advocacy group in New York, said targeting the network was justified because ?SNAP is a menace to the Catholic Church.?

Mr. Donohue said leading bishops he knew had resolved to fight back more aggressively against the group: ?The bishops have come together collectively. I can?t give you the names, but there?s a growing consensus on the part of the bishops that they had better toughen up and go out and buy some good lawyers to get tough. We don?t need altar boys.?

He said bishops were also rethinking their approach of paying large settlements to groups of victims. ?The church has been too quick to write a check, and I think they?ve realized it would be a lot less expensive in the long run if we fought them one by one,? Mr. Donohue said.

However, a spokeswoman for the United States Conference of Catholic Bishops, Sister Mary Ann Walsh, said Mr. Donohue was incorrect.

?There is no national strategy,? she said, and there was no meeting where legal counsel for the bishops decided to get more aggressive.

Mr. Clohessy and others founded the survivors network as a loose collective of volunteers who had been victimized by Catholic priests. Their goal was to help others grapple with the emotional and psychological fallout. They make referrals to therapists and lawyers, and hold protests outside church offices.
